IBA Lifesciences 社（以下 IBA 社）独自の Strep-tag® / Strep-Tactin® システムは、ビオチン-ストレプトアビジンの相互作用を用いたアフィニティクロマトグラフィーシステムで、His-Tag よりも高純度に精製が可能です。第 3 世代の Twin-Strep-tag® / Strep-Tactin® XT は、従来と比べて結合親和力が高く、共有結合に近いことから、より高い収率でタンパク質を得ることができます。

本システムは、

  • 2 つのアフィニティタグ（Strep-tag® II、 Twin-Strep-tag®）
  • 2 つのストレプトアビジン誘導体（Strep-Tactin®、 Strep-Tactin® XT）

で構成されており、Strep-tag® 融合タンパク質の精製に使用します。

Strep-tag® II の Strep-Tactin® への結合親和性（1 µM）は、ストレプトアビジンへの結合親和性のほぼ 100 倍です。さらに改良された Strep-Tactin® XT は、Strep-tag® II に対して nM の親和性、Twin-Strep-tag® に対して pM の親和性を持っています。

GFP の 6xHis-tag（A）または Twin-Strep-tag®（B）融合タンパク質を、大腸菌のライセートから分離させた。His-tag には Ni-NTA（サーモフィッシャーサイエンティフィック）、Twin-Strep-tag® には Strep-Tactin® XT 4Flow high capacity（IBA Lifesciences）を使用した。
