～ Unravel the Mysteries of STING ～
STING（stimulator of interferon genes）は、小さな核酸分子である環状ジヌクレオチド（CDNs）により活性化され、I 型インターフェロンや炎症性サイトカインを誘導することが知られています。STING リガンドには、細菌が感染した際に形成される CDNs や細胞質 DNA を認識する cGAS により合成される内因性の CDN、2'3'-cGAMP があります。
本セミナーでは、STING の機能について最新の論文情報を交えて紹介します。STING と他の核酸センサーおよびアダプターとの相互作用や、感染症、自然免疫、がんおよびワクチンアジュバント研究との関係性について説明します。次に、がん治療を目的に、合成した CDNs を用いて免疫応答を惹起させ、がん細胞の細胞死および抗原提示細胞の活性化を試みました。ここでは、STING をターゲットとした CDNs のライブラリーをデザインし、その結果判明した、免疫チェックポイント阻害剤とのシナジー効果をもつ CL592 および CL656 の強力な抗腫瘍特性について説明します。また STING の研究に有用な製品群も紹介します。
