Mca-APK(Dnp) ACE2 substrate is a FRET peptide for measuring enzymatic activity in cells and tissues.
This fluorogenic substrate offers more flexibility and higher throughput than the commonly used HPLC-separation-based methods.

The spike protein of SARS-CoV-2 utilizes the ACE2 surface receptor on host cells to promote cellular entry, making it a promising therapeutic target for SARS-CoV-2. The loss of ACE2 function following binding by SARS-CoV-2 is driven by endocytosis and activation of proteolytic cleavage and processing.

To screen and evaluate ACE2 inhibitors AAT Bioquest offers Mca-APK(Dnp), a fluorogenic ACE2 substrate:

 

  • The substrate has an ex/em maxima of 322 nm and 381 nm, respectively
  • Uses DNP as a quencher molecule to quench the fluorescence of methoxycoumarin (Mca).—more flexibility and higher throughput than the commonly used HPLC-separation-based methods
  • Suitable for either kinetic or end-point reading, depending on protocol.
