Glutathione Dose Response Measurements: Assay Signal Consistency & Sensitivity
In healthy cells and tissues, glutathione exists in two states: reduced (GSH) and oxidized (GSSG). In the reduced state, GSH is a major tissue antioxidant that neutralizes reactive oxygen species (ROS). Under normal conditions, healthy mammalian cells typically have molar GSH/GSSG ratios exceeding 100:1. Under oxidative stress however, GSSG accumulates and the ratio of GSH/GSSG decreases to ratios as low as 10:1 and even 1:1. As such, monitoring GSH/GSSG levels has become a useful tool in evaluating oxidative stress in cells and tissues. AAT Bioquests' kits provide a sensitive, one-step fluorimetric method to detect as little as 1 picomole of cysteine or GSH in a 100 µL assay volume, and can be read by a fluorescence microplate reader and easily adapted to automation without a separation step. |
