Membrane Protein Structure Determination in 2019 was impressive. Want to know how it was done?

 

Back by popular demand is our annual summary of membrane protein structural biology from the previous year. Using data from Steve White’s “Membrane Proteins of Known 3D Structure” and our very own Detergent Database, we reveal information regarding the methods and detergents used for membrane protein structure determination in 2019. And with 122 unique membrane structures determined, it was not only a record-breaking year for membrane protein structural biology, but the results make for a very interesting read.

For example, we’re still witnessing the tremendous impact Cryo-EM is having on membrane protein structural biology, but X-ray crystallography is not going anywhere. 2019 saw a nearly 50/50 split between the structures determined using Cryo-EM and X-ray crystallography. And DDM continued to be the most commonly used detergent for membrane protein solubilization and purification. However, what’s interesting to us is that 2019 saw more structures determined using GDN to Digitonin for the first time ever – which demonstrates that the membrane protein community has adopted GDN as an effective synthetic drop-in substitute for Digitonin. Adding to our excitement is the first structure determined using the trehaloside detergent, 6-DDTre.
