LifeSensors and the New Developments in the Ubiquitin Proteasome System
The ubiquitylation reaction is a sequential process mediated by three enzymes: E1, E2, and E3.
LifeSensors’ TUBEs
●LifeSensors has developed TUBEs (Tandem Ubiquitin Binding Entities) for detecting and isolating poly-ubiquitin proteins.
●We have developed Pan Selective TUBEs and poly-ubiquitin chain selective TUBEs lysine 48, lysine 63, and Linear/M1.
●Applications:
○Enrichment of poly-ubiquitylated proteins
○Monitoring the formation of new poly-ubiquitin chains as part of an E3 ligase assay
○Detection of poly-ubiquitylated proteins by western blots in a more efficient manner than antibodies
○Imaging of cells, organelles, or tissue
TUBEs and PROTAC Discovery
●PROTAC (Protein Targeting Chimera) molecules can be used to hijack the Ubiquitin Proteasome System to degrade disease associated proteins.
●TUBEs allow for the detection of the poly-ubiquitylated protein prior to its degradation, thus establishing the PROTAC drug function.
●LifeSensors has developed plate-based High-Throughput Screening (HTS) methods to detect PROTAC ubiquitylation.
●Advantages of LifeSensors' technologies over others available on the market:
○Provide more information on the state of the protein
○Only PROTAC targeted assay that can establish poly-ubiquitination and subsequent degradation in rapid HTS fashion
○Do not require establishing stable cell lines
○Allow for testing in more physiologically relevant conditions
○Poly-ubiquitin chain selective platforms to decode complex PROTAC mechanisms
